In vivo study | In macaques, Naltrexone (0.32 mg/kg) attenuated the ethanol-enhanced response at the concentration that maintained the highest response (1% or 2%). Naltrexone (0.1 mg/kg) at a low ethanol concentration (0.25%) yielding a small ethanol uptake (g/kg) and at a high ethanol concentration (4%) yielding a modest amount of uptake weakens the ethanol-enhanced response. Naltrexone (1-3 mg/kg) potently and dose-dependently inhibited the ethanol demand generated by the non-accidental delivery of 8% ethanol-filled liquid scoops. When Naltrexone (approximately 100 ng/kg) was co-administered with morphine (3 mg/kg), the analgesic efficacy of morphine was optimally amplified. In the rat tail-flick test, Naltrexone (10 ng/kg I. p.) enhanced morphine acute submaximal dose intracapsular (5 mg) or systemic (7.5 mg/kg I. p.) analgesic effect of drug administration. In rats, Naltrexone in combination with morphine inhibits the attenuation of morphine analgesia and prevents the loss of morphine efficacy. Naltrexone significantly inhibited ethanol self-administration and prevented the ethanol-induced increase in osmotic dopamine levels. Naltrexone completely prevented the reduction of the distance between the anus and the genitals in prenatal stress (PS) males and restored the growth rate of both sexes. Naltrexone also reduced anxiety in PS rats in the cross maze, increasing the measured opioid component to control levels, but increasing anxiety in control men. |
